Yes.
Squid is an application level HTTP proxy. The protocol used to talk
to the proxy is HTTP. Sits at the same ISO/OSI level as browser or
HTTP server applications.
Further to narrow down the scope, the networking interface of Squid
expects HTTP to be running on top of IPv4. (IPv6 is in development)
